abstract The present invention relates to a method of quantifying DNA strand breaks in intact cells whereby (a) cells are lysed in order to release DNA, (b) the lysate is subjected to an alkaline denaturation process, (c) neutralisation occurs, (d) a fluorescent dye is added and (e) fluorescence is read off an fluorometer. The method is characterized in that all pipetting stages are carried out automatically by means of a pipetting device in a lightproof housing. The invention further relates to a device to carry out said method, comprising a lightproof housing, a temperature controlled base plate, a system of pipetting nozzles and a fluorometer to measure the fluorescent intensity of DNA.
