abstract |
There is described a human proinsulin derivative of the formula: human insulin B chain - Z - human insulin A chain wherein Z is a polypeptide linker of the formula: U-U-X-P-J-(X')n-U-U in which: U is arginyl or lysyl; X and X' are amino acid radicals; P is prolyl; J is glycyl, arginyl or lysyl; n is 0 or an integer of from 1 to 5. There is also described a DNA sequence encoding the proinsulin derivative, which may be inserted into an expression vector. The vector is used to transform a microorganism, preferably Escherichia coli . Human insulin is prepared from the proinsulin derivative by enzymatic hydrolysis. |
