abstract | Pharmaceutical preparation consists of a sterile, injectable, physiologically acceptable solution which releases nitric oxide, NO, free from oxygen, on increase in temperature. The solution comprises a 0.9% sodium chloride solution which is saturated with NO. Also claimed is a process for quantitative determination of endothelial dysfunction in mammals. The process comprises (a) transferring a measured volume of whole blood into an equal volume of 1M NaOH as a stop solution, under conditions which exclude oxygen; (b) adding 50 mu l 1M phosphoric acid and centrifuging the mixture for 5 minutes at 10000g; (c) decanting the supernatant into an ultrafiltration tube and centrifuging for 15 minutes at 2000g; (d) diluting the ultrafiltrate with HPLC-water (1:10); and determining the nitrite content by known methods. - |
