abstract | Purification of anti-A-hel agglutinin from Helix pomatia Crude extract from dried or freshly pulverized protein glands or viscera of Helix pomatia (obtainable by extracting with physiological saline, opt. concentrating by (NH4)2SO4 precipitation, dialysing salt-free against dist. H2O and lyophilizing) is adsorbed on 'Sephadex' (pref. G 200) and the agglutinin is eluted stepwise with phosphate buffer (pref. M/20, pref. pH 7.0) and the same buffer to which dextrose has been added (pref. in a concn. of 0.5-M); the product is freed from salts by dialysis, then lyophilized. Process can be used on technical scale and yields as product free from ballast substances. Residues from production of edible-snail preserves can be used as starting material. Anti-Ahel agglutinin is a glycoprotein which can be used to specifically detect sub-group H A1 blood corpuscles as well as group C Streptococci. |
