abstract |
Provided herein is a method for chemically capping polynucleotides with a 5'-monophosphate. In some embodiments, the method may include: combining an activated 5'-nucleoside mono- or polyphosphate with a polynucleotide population comprising polynucleotides having a 5'-monophosphate to produce a reaction mixture; and incubating the reaction mixture to produce reaction products comprising a polynucleotide and a 5' nucleoside cap linked by a 5'-5' polyphosphate bond. The chemical capping method described here can be integrated into a variety of cDNA synthesis methods. |
