Described are nucleic acid hybridization buffer formulations and uses thereof that provide improvements in hybridization specificity, rate, and efficiency. The buffer formulation composition contains a target nucleic acid, at least one polar, aprotic, organic solvent and a pH buffer system, wherein the target nucleic acid is attached by hybridization with a surface-bound nucleic acid which is bound to the surface, and wherein the hybridization of the target nucleic acid and the surface-bound nucleic acid has a high stringency and melting rate. |
