abstract | The present invention is based on the idea of simulating a wound extracorporeally in order to obtain wound healing / regenerative growth factor mixtures. The invention provides a method of separating the growth factors released from cells after coagulation (coagulation-induced signal phase) from the growth factors secreted by cells under hypoxia (hypoxia-induced signal phase), followed by mixing the two phases at defined ratios to get a new composition. Peripheral blood is collected and coagulated or centrifuged to induce coagulation. The serum is completely or partially removed and replaced with fresh medium with a volume equal to or less than the volume of the serum removed. The clot is incubated in fresh medium at a temperature of 10 to 40 ° C under hypoxia (1 to 10% O 2 ) for 1 to 7 days. The clot-conditioned medium is then collected and mixed with the serum in a defined ratio to obtain the composition. Compositions comprising adjustable concentrations of growth factors derived from blood serum (coagulation-induced phase) and clot-conditioned medium (hypoxia-induced phase) can be added to suitable carriers and can be applied topically or injected into a subject to aid wound healing, skin regeneration or rejuvenation support. |
