abstract The present invention relates to a method of light-directed generation of covalent conjugates between each two molecules of the same or different chemical nature by a light-controlled addition reaction, preferably a 2-2-cycloaddition, for the covalent connection between two identical or different functional groups selected from the functional groups maleimide derivatives, maleic acid derivatives, maleic acid monoamide derivatives and maleic acid diamide derivatives and their 2- and / or 3-substituted derivatives. According to the invention, the addition reaction takes place as a two-photon reaction at a wavelength in the range of λ = 600 to 900 nm. Thus, a light-driven reaction for spatially and temporally defined generation of strong covalent bond between two molecules is disclosed without the need for the use of a photoinitiator. The method can be used to form site-specific conjugates with favorable pharmacological properties, for example, reduced immunogenicity and increased circulation time. The described method can be used for molecular conjugation in a solution of any kind. In particular, the lack of photoinitiator and the low energy allows the application of the method in vivo, which in turn allows in situ application for two-dimensional and three-dimensional structuring, manipulation and structuring in the presence of living cells.
