abstract | The invention relates to a process for the preparation of drospirenone starting from 15,16-methylenprasterone by reaction with a strong base and subsequent reaction of the reaction product of lithium or butyllithium and 1-bromopropyl-3-trimethylsilyl ether to 15,16-methylene-17β-hydroxy 17α- (propyl-3-trimethylsilyloxy) -prasterone, cleavage of the trimethylsilyl protecting group and oxidative ring closure to 15,16-methylene-17 (S) -spirolactone-androst-4-en-3-one (MADO-lactone); Enolization to 3-methoxy-15,16-methylenandrost-4,6-dien-3-one-17 (S) -spirolactone, subsequent oxidation to 15,16-methylenandrost-4,6-dien-3-one-17 ( S) -spirolactone (MDD-lactone) and methylenation to drospirenone. Furthermore, the invention relates to a synthesis route from Prasteron drospirenone on the above reaction sequence. |
