abstract | Method for the analysis of cytosine methylations using DNA repair enzymes, characterized in that: a) the DNA to be examined is reacted chemically or enzymatically such that 5-methylcytosine remains unchanged, while unmethylated cytosine is converted into uracil or into another base, which differs in the base-pairing behavior of cytosine, b) the transformed DNA is hybridized with oligonucleotides, wherein the DNA of one methylation state forms hybrids with base mismatches, while the DNA of the other methylation state forms hybrids without base mismatches or no hybrids, c) one strand of the mismatched hybrids is enzymatically cut, d) the uncut DNA or the cut fragments are detected, e) from the signal on the methylation status of the examined DNA is concluded. |
