abstract | Method for analyzing cytosine methylation patterns in genomic DNA samples, comprising the following steps: a) the genomic DNA is isolated from cells or other accompanying materials and bound substantially irreversibly to a surface, b) the DNA bound to the surface is treated, preferably with a bisulfite, such that cytosine is converted into a base which is different from the base pairing behavior in the DNA duplex, while 5-methylcytosine remains unchanged, c) the reagents used in step b) are removed in a washing step, d) selected portions of the immobilized DNA are amplified in a polymerase reaction, e) the amplificates are examined with regard to their sequence. |
