abstract A method of identifying a region of a living cell genome to which a protein of interest binds, comprising the steps of: a) linking DNA-binding protein in the living cell with genomic DNA of the living cell, wherein genomic DNA cross-linked, DNA-binding protein is produced; b) generating DNA fragments of the DNA-binding protein-associated genomic DNA in a), wherein a mixture is generated with DNA fragments to which DNA-binding protein is bound; c) removing a DNA fragment to which a protein of interest is bound from the mixture prepared in b); d) separating the DNA fragment identified in c) from the protein of interest; e) amplifying the DNA fragment of d) using a polymerase chain reaction; f) contacting the DNA fragment of e) with DNA having a sequence complementary to genomic DNA of the cell under conditions where hybridization between the DNA fragment and a region of genomic DNA is present.
