abstract The invention relates to a process for the complete restriction of DNA molecules. The invention relates to the field of genetic engineering, where a specific fragmentation of DNA molecules by means of restriction endonucleases belongs to the basic methods. The object of the invention is to completely and rapidly break down DNA molecules which were only incompletely cleavable using certain previously known restriction endonucleases using the previously known method into specific restriction fragments. The object was achieved by incubating such DNA molecules with the restriction endonuclease in the presence of a second DNA species (in particular oligonucleotide duplexes which contain the recognition site for the corresponding restriction endonuclease). The method for completing and accelerating the specific cleavage of phage lambda DNA by the restriction endonucleases Eco57I and Cfr9I, DNA of the phage vector mp18 by EcoRII and the plasmid pBR322 by NarI proved to be very advantageous. {Genetic engineering; restriction endonucleases; DNA; DNA cleavage; oligonucleotides; phage; vectors; plasmids; Cfr9I}
