abstract | An operator of the E. coli phage lambda cIts repressor is modified so that there is a consensus sequence between the two binding sites of the repressor molecules for a TATA box (especially the yeast), but the binding affinity of the repressor to the so modified operator is not may change. This modified operator is incorporated in place of or downstream of the natural TATA box or functionally similar regions of any eukaryotic promoter and transferred into a eukaryotic host previously altered to produce cIts repressor molecules by means of a vector. The original transcriptional strength of the eukaryotic promoter is not impaired. Depending on the synthesis of cIts repressor molecules, transcription of the eukaryotic promoter is repressed with the modified operator.
