abstract | The invention relates to a genetic-microbial method for obtaining authentic serotype C streptokinase. It aims to effectively produce the said protein. The task associated with this objective is achieved as follows: First, a cell population of one strain of the species Streptococcus equisimilis is transformed with a recombinant plasmid vector replicable in Gram-positive bacteria which, in addition to the coding DNA, encodes the coding DNA sequence for at least one phenotypic marker that can be expressed in Streptococcus authentic serotype C streptokinase carries. Transformed recipient cells obtained here are subsequently amplified for 8 to 20 generations without selection pressure with respect to the plasmid-encoded phenotype marker and checked for the combination of features plasmid-free and simultaneous preservation of said marker. As a result of this procedure, homologous recombination produces mutant S. equisimilis strains in which the skc gene is duplicated in the chromosome of the host.
