titleOfInvention | PROCESS FOR PREPARING ALPHA ERGOKRYPTIN AND ERGOSINE |
abstract | To provide alpha-ergokryptine and ergosine in sufficient quantities for therapy and for partial synthetic work, the task was to produce these alkaloids by saprophytic culture of a fungus strain. The task was solved by using the strain Claviceps purpurea (Fr.) Tul. To produce alpha-ergocryptine and ergosin. IBP 84, ZIMET 43 768, or one of its mutants or variants. The strain is cultured in aerobic submerged fermentation using nutrient media containing assimilable carbon, assimilable nitrogen sources and mineral salts at a temperature of 20-30C and a pH of 4.0-6.4. After 7-14 days of main culture, the strain forms a total alkaloid mixture of 700-1,400 g / ml of culture suspension of which 200-400 g / ml of culture suspension are cyclolalkoloids. At least 90% of the alkaloids are isolatable from the culture filtrate. The resulting cyclolalkaloids are alpha-ergokryptin / alpha-ergokryptinin (80%) and ergosine / ergosinine (20%). The ratio is controllable by Zufuetterung of the specific Cyclolteil specific amino acids, valine or alanine. |
