abstract | The invention relates to a process for the preparation of substituted 3-cyano-pyrid-2 (1H) -ones with optionally cycloaliphatic cross-linking in the 4,5- or 5,6-position. It is the object of the invention to represent these compounds in a simple manner with good yields. The task is to find a technically feasible synthetic route. The object is achieved by the reaction of substituted 3-cyano-pyrid-2 (1H) thiones with 2-bromoethanol to give the 2- (2-hydroxyethylthio) pyridine-3-carbonitriles. Subsequent heating of these compounds with alkali metal alkoxides in alkanols (C1C3) or by means of diluted alkaline solutions in conventional protic or aprotic solvents, in each case under reflux, gives the 3-cyano-2 (1H) -pyridones in very good yields. The 3-cyano-2 (1H) -pyridones can be substituted in different ways in the 4- and 6-positions or in cycloaliphatic bridged positions in 4,5- or 5,6-position. The invention has significance for the pharmaceutical and chemical industries. |
