abstract | The method finds application in the preparation of DNA fragments from natural DNA and their use as primers in cDNA synthesis and oligo-labeling. The method allows the preparation of the primer DNA without elaborate fractionation techniques. The essence is that the DNA is digested sequentially with two endonucleases (DNase I and micrococcal nuclease), with DNase I treatment in Mn-containing solution. As a result, fragments are obtained which show virtually no self-installation of radioactivity during the synthesis reactions. Additional fractionation is achieved by differential ethanol precipitation. |
