abstract | The invention relates to a method which is suitable for the preparation of a novel expression vector based on a hybrid promoter. The invention has for its object to provide a new principle solution for the effective expression of eukaryotic genes and thus the economically viable biosynthesis of animal proteins in microorganisms, especially in E. coli. According to the invention, the coding DNA sequence of a polypeptide, in particular human pregroinsulin, human interferon, BLV proteins, influenza virus proteins or partial sequences of these proteins is linked to a unit containing the nucleotide sequence segments necessary for the control of the expression such that the control of the Expression a combined unit is used which contains nucleotide sequence sections of at least two different natural, prokaryotic control units. A combined unit is used which is unique in terms of base sequence and distance between the major components essential for transcription and translation, particularly "-35-, 10-region", transcription start point, 16 S-RNA binding site and translation start point, by in vitro mutagenesis varies. |
