titleOfInvention MERIDIFFUSION ANALYSIS FOR THE DETERMINATION OF ENZYMIN HIBITORS
abstract The object of the invention is the semi-quantitative determination of an inhibitor which is in a mixture with other inhibitors, but which have a higher molecular weight than the latter. The object of the invention is to modify the known geldiffusionsanalytische semiquantitative determination of inhibitors so that it is possible to detect low molecular weight inhibitors even if they are present in admixture with higher molecular weight inhibitors. In the conventional method, holes are punched into a gel into which the samples to be tested for enzyme inhibitors are pipetted. Since the gel simultaneously contains an enzyme substrate, it can be checked whether the samples to be tested have an inhibitory effect on the degradation of the enzyme substrate contained in the gel by an enzyme. According to the invention, the object has been achieved by pipetting the inhibitor mixtures onto the gel-deposited membranes which contain higher-molecular-weight enzyme inhibitors, whereby only the low-molecular-weight inhibitors diffusing through the membrane are detected. The exclusion limit of the membrane must be selected according to the molecular weights of the inhibitor mixtures.
