METHOD OF GENERATING A RECOMBINANT DNA CONTAINING AN AMYLASE-CODING GENE |
abstract |
The invention relates to a novel recombinant DNA containing an amylase-encoding gene and produced by an in vitro method in which the DNA derived from a bacterial donor microorganism is split and the resulting DNA fragments are linked to a vector which is similarly linked has been split, characterized in that the vector is a plasmid or the DNA of a phage lambda derivative. The invention further relates to genetically engineered microorganisms containing the novel DNA, to methods of producing the novel DNA and to novel amylase enzymes, their production from DNA and their use for the hydrolysis of starch. The donor microorganism is preferably a Bacillus or Klebsiella strain. The vector may be a phage or a plasmid. The host microorganism is preferably E. coli or B. subtilis. The produced amylase enzyme is preferably an alpha-amylase, a beta-amylase or a pullulanase. |
