Plasmids for use in recovery of a plasminogen activator (PA) have an operon consisting of a regulatable promoter; an active Shine-Dalgarno site (SDS) as ribosome-binding site; a start codon; a synthetic gene (I) for the 411 amino acid, single-chain urine-PA 'SCU-PA' and, downstream of the gene, 1 or 2 terminators. The plasmids are suitable for expressing recombinant SCU-PA precursor (II) in Enterobacteriacae, esp. E. coli. The distance between SDS and the start codon is 6-12 (esp. 8-10) nucleotides, and the promoter is the Trp or Tac promoter. The terminators are trpA and/or the tetA orfL terminator from Tn10. |
