abstract | The method described allows two-stranded DNA to be effectively labeled. The method consists in incubating double-stranded DNA in the presence of monovalent copper and oxygen ions and / or active oxidants. The source of these ions may be aqueous solutions containing copper ions, or may be a monovalent honey that has been bound to the DNA. Due to the action of oxygen and monovalent copper ions, DNA strands result in breaks that serve as the starting point for DNA strand synthesis using these enzymes in the subsequent step of incubation with labeled nucleotides and enzymes with DNA polymerase activity. Either terminal deoxynucleotidyl transferase, or DNA polymerase, synthesizing the novel chain according to the master strand may be used. If these DNA polymerases do not have exonuclease activity and / or displacement activity, it is necessary to incubate samples containing double stranded DNA with enzymes having exonuclease and / or phosphatase activity prior to incubation with DNA polymerase to ensure sufficient signal strength. The incubation step with these enzymes can also be used to increase the signal when subsequent DNA polymerases with exonuclease and / or displacement activity are used. In the case of terminal deoxynucleotidyltransferase, it is advantageous to incubate in the presence of an enzyme with 3'-5 'exonuclease and / or phosphatase activity prior to incubation with this enzyme. Rinsing agents may optionally be used to remove unwanted copper products. |
