abstract | The present invention relates to a new process for the preparation of (-) - 1- (3-hydroxypropyl) -5 - [(2R) -2 - ((2,2,2-trifluoroethoxy) phenoxyethyl} amino) propyl] -2,3-dihydro The 1 H -indole-7-carboxamide of formula I, also known as silodosin, and its salts, proceeding in 4 steps: step A is alkylation of a secondary amine of formula II, an alkylating agent of formula III, step B is N-hydrogenolysis the tertiary amine of formula IV with hydrogen on a metal catalyst, step C is the hydrolysis of the nitrile of formula V by treatment with alkali, and step D is the O-debenzylation of the amide ether of formula VI with hydrogenolytic or trialkylsilyl halide reagents. |
