abstract In order to obtain structural and functional information about proteins, based on polarizing fluorescence microscopy, a fluorophore-bound protein is subjected to two-photon or multi-photon fluorescence microscopy, whereby the investigated protein with the fluorophore is irradiated with a laser beam whose light is polarized at at least two different polarizations and which is excites the fluorescence of the fluorophore, and based on the localization, intensity, and polarization of fluorescence excited by the different polarizations of the excitation laser, the localization and extent of anisotropy of absorption and / or localization and the degree of anisotropy of the fluorescence are determined to determine the structural and functional and properties of the protein . A suitable device for obtaining structural and functional information on polarizing fluorescence microscopy proteins comprises a modulator (P) for rapidly modulating the polarization of the excitation beam (1) to produce two-photon or multi-photon fluorescence and a control unit (R), wherein the modulator (P) and the control function. unit (R) is synchronized with microscope scanning (M) such that the fluorescence intensity information recorded by the microscope (M) is assignable to the different polarization states of the excitation beam (1) based on the time profile of the modulator of the excitation beam polarization polarization (1) used (P).
