The method of preparation comprises reacting a tetracycline chemical complex containing a tetracycline-based reactive precursor having at least one reactive site selected from 7-, 9- or 13- and a transition metal catalyst to form a reactive chemical intermediate with a reactive organic substituent precursor or a reactive organic combination. a tetracycline-based precursor having at least one reactive site selected from position 7, 9 or 13 and a reactive organic substituent precursor in the presence of a transition metal catalyst under conditions whereby a tetracycline compound substituted by said organic substituent is formed, wherein said tetracycline compound is, e.g., oxytetracycline, chlortetracycline, demeclocycline, etc., and wherein the tetracycline compound is substituted with one or more substituents including, for example, alkyl, alkenyl, alkynyl, cycloalkyl, etc. Rea a tetracycline chemical complex comprising a tetracycline-based reactive precursor and a transition metal catalyst.
