Avian paramyxovirus cDNA comprising at least a nucleic acid sequence corresponding to the 5 'end of the Newcastle disease virus genome as seen in Figure 6, allowing the preparation of an infectious copy of avian paramyxovirus. A method for preparing an infectious Newcastle disease virus (NDV) by cloning full-length cDNA and using vaccines and diagnostic assays developed and derived during the preparation of an infectious NDV particle. The method offers the possibility of modifying the NDV genome by means of genetic modifications and allowing the introduction of mutations, deletions and / or insertions. The method makes it possible to prepare live NDV marker vaccines that can be serologically distinguished from NDV field strains. |
