abstract | A method for preparing monomeric conjugates of calicheamicin derivative and carrier with active ingredient content, high yield and reduced agglomeration. These conjugates are prepared by incubating a calicheamicin derivative and a protein carrier in a non-nucleophilic, protein-compatible, buffered solution having a suitable pH ranging from 4.0 to 8.5, and containing a co-solvent selected from the group consisting of propylene glycol, ethanol, DMSO and combinations thereof and an additive comprising at least one carboxylic acid of from 6 to 18 carbon atoms, at a temperature in the range of about 25 to 37 [deg.] C. and about 15 minutes to 24 hours; and purifying the conjugate prepared in step (1) to form monomeric conjugates. Alternatively, the conjugates can be prepared by incubating the calicheamicin derivative and the protein carrier in a solution containing non-nucleophilic, protein-compatible, buffered solution and co-solvent containing t-butanol. |
