abstract | Recombinant heat stable DNA polymerase, which is a mutant form of a naturally occurring thermostable DNA polymerase, wherein the naturally occurring heat stable DNA polymerase has an amino acid sequence comprising the amino acid sequence motif SerGlnIleGluLeuArgXaa (SEQ ID NO: 2), wherein Xaa at position 7 of this sequence motif is Val or Ile, and the mutant form is modified to contain a non-Glu amino acid at position 4 of the sequence motif, and the mutant form has reduced discrimination against unusual nucleotide incorporation as compared to the naturally occurring thermostable DNA polymerase. Polymerase has improved efficiency in incorporating unusual nucleotides such as ribonucleotides into DNA products and is useful in many in vitro synthesis applications. It is particularly useful for nucleic acid sequence analysis protocols and provides new means for DNA sequence analysis with cost and efficiency advantages. Nucleic acids encoding such polymerases, vectors and host cells containing such nucleic acids, and compositions for use in DNA sequencing reactions, kits, and sequencing methods using such polymerases. |
