abstract |
The present invention relates to a method for producing tryptophan-producing microorganisms, wherein a micro-organism that is deregulated in at least one tryptophan biosynthesis step is provided with a serA allele having reduced serine inhibition, wherein microorganisms that are deregulated in at least one tryptophan biosynthesis step, are subjected to mutagenesis and the mutagenized population of organisms is selected for clones with serA genes that encode serine insensitive phosphoglycerate dehydrogenases or ser alleles that encode serine insensitive phosphoglycerate dehydrogenases are produced by genetic engineering techniques, preferably by site-directed mutagenesis techniques and such alleles are introduced into microorganisms that are deregulated in at least one tryptophan biosynthesis step such that they are integrated either on an autonomously replicable recombinant vector or on the strain chromosome. It further relates to the use of the microorganisms thus produced for the fermentative production of tryptophan |
