abstract The present invention provides a method for determining the deaminase activity of cytosine derivatives converting enzymes to uracil derivatives in a sample by incubating the sample with an enzyme substrate selected from the group consisting of 5-ethynyl-2'-deoxycytidine (EdC), 5-bromo-2'-deoxycytidine (BrdC) , 5-chloro-2'-deoxycytidine (CldC), 5-iodo-2'-deoxycytidine (IdC), 5-ethynylcytidine (EC), 5-bromo-cytidine (BrC), 5-chloro-cytidine (ClC), 5-iodo-cytidine ( IC) and 5-fluorocytidine (FC), these substrates being converted to deaminated products in the presence of cytosine derivatives to uracil derivatives, EdC is converted to 5-ethynyl-2'-deoxyuridine (EdU), BrdC to 5 -bromo-2'-deoxyuridine (BrdU), CldC to 5-chloro-2'-deoxyuridine (CldU), IdC to 5-iodo-2'-deoxyuridine (IdU), EC to 5-ethynyluridine (EU), BrC to 5-bromuridine (BrU), ClC to 5-chloruridine (ClU), IC to 5-ioduridine (IU) and FC to 5-floururidine (FU), these deaminated products are subsequently detected in DNA or R NA and determined in the amount of incorporated EdU, BrdU, CldU, respectively. IdU in DNA and EU, BrU, ClU, IU, resp. FU in RNA. The present invention also includes the use of the aforementioned method for determining deaminase activity in cells treated with substances that affect the deaminase activity of cytosine derivative converting enzymes to uracil derivatives.
