abstract | The present invention relates to a recombinant soluble T receptorncells. The T cell receptor (TCR) is folded into the correct onenspatial arrangement and contains recombinantnthe extracellular domain of α or g of the TCR chain having the firstnheterologous C-terminal dimerization peptide, and recombinantnthe extracellular domain of b or d of the TCR chain having a secondna heterologous C-terminal dimerization peptide that is specificallynheterodimerized with the first dimerization peptide, songenerates a heterodimerization domain, which may be a domainnwith the coil coil conformation. The solution further provides sequencesnnucleic acids encoding recombinant TCR and methodnpreparing recombinant TCRs. TCRs may be labeledna detectable label so that they can detect specificnMLIC-peptide complexes. Alternatively, they may benlinked to a teape agent such as cytotoxicnor immunostimulating the agent to transfer the agent toninstead of a specific MHC-peptide complex. |
