abstract | Preparations for parenteral administration of certain hydrophobic active agents such as Coenzyme Q10 are disclosed herein. Methods are also provided herein and methods for treating oncological disorders by the use of the same. The preparations contain an aqueous solution • a hydrophobic active agent which is dispersed to form a colloidal nano-dispersion of particles; and at least one of a dispersion stabilizing agent and an opsonizing agent, where the colloidal nano-dispersant is into nano-dispersion particles having an average size of less than 200 nm. Methods of preparing parenteral preparations include dispersing the hydrophobic active agent by high pressure homogenization by (1) adding the hydrophobic active agent to a 65 ° C water bath and mixing to form a hydrophobic active agent / water additive • (2) additive disperse into the hydrophobic active agent / water mixture and mix at 65 ° C to form a hydrophobic active agent / water / stabilizing agent mixture (3) adding an agent a reduction of opsonization to form a mixture of hydrophobic active agent / water / stabilizing agent / reducing agent; (4) reheating a microfluidizer at 65 ° C; and (5) treating by mixing the hydrophobic active agent / water / stabilizing agent reducing agent in the microfluidizer at 65 ° C to form a colloidal nano-dispersion of the hydrophobic active agent having an average particle size of less than 200 nm. Methods for treating oncological disorders are also provided herein by administering the preparations described herein to a subject in order to treat or prevent the oncological disorder. |
