abstract A stable high temperature and high purity crosslinked (optionally> 70% β-β bound) tetrameric hemoglobin with high oxygen delivery efficiency suitable for use in mammals without causing renal injury and vasoconstriction is provided. The bilateral form of hemoglobin is degenerated and purification procedures are performed on whole blood red blood cells. Controlled hypotonic lysis in a direct cytolysis device prevents white blood cell lysis. White blood cell nucleic acids and phospholipid impurities are not detected. Blocking of the reactive sulfhydryl groups with a sulfhydryl reagent is carried out in an oxygenated environment. Cross-flow column chromatography removes different plasma protein impurities. N-acetyl cysteine is added to the cross-linked tetrameric hemoglobin to maintain a low level of meta-hemoglobin. The stabilized hemoglobin is maintained in an aluminum wrapper infusion bag to prevent the formation of inactive met-hemoglobin from oxygen intrusion. The product is used in tissue oxygenation and cancer treatment.
