abstract <p> The invention provides a method for the formation of a thorium complex directed to tissue, wherein said method comprises; <br /> a) forming an octadentant chelator comprising four hydroxypyridinone (HOPO) moieties substituted at position N with a C1-C3 alkyl group, and a coupling portion ending in a carboxylic acid group; <br /> b) coupling said octadentant chelator to at least one peptide or protein targeting to <br /> tissue comprising at least an amine portion by means of at least one amide coupling reagent to thereby generate a tissue targeting chelator; and <br /> c) contacting said tissue targeting chelator with an aqueous solution comprising an ion of at least one alpha-emitting thorium isotope. <br /> A method of treating a neoplastic or hyperplastic disease that it comprises the administration of said thorium complex directed to tissue, as well as the corresponding pharmaceutical complex and formulations. </p>
