titleOfInvention | A method for isolating immunoglobulin by affinity chromatography |
abstract | The solution relates to the method of plasma isolationnimmunoglobulin by affinity chromatographynon substituted porous onesnspherical celluloses with immobilizednligand of histidine, lysine, reactivenSluti 13 (C.I. 18,990) or 2- (4-aminophenyl) sulfonylethyl groupsnand productsntheir diazotization and spontaneous transformation.nThe principle of a procedure that is based on non-specificnimmunoaffinity marked functionalngroups to immunoglobulin is basednon its sorption, separation of unattached proteinsnand subsequent desorption with concentratednwith a solution of salts, preferably 3.5M chloridenof magnesium or calcium. To increaseneffect of 1,4-dioxane is recommended, noptionally stabilizing components such asnsodium acetate, glucose or maltose. Acquirednthe eluate is freed from excess saltndialysis or diafiltration. In the above mannernit is possible to prepare fully nativenelectrophoretically pure immunoglobulinnigG. |
