abstract 1. The present invention describes methods and processes for the production of proteins, particularly glycoproteins, by animal cells or mammalian cell cultures, illustratively, but not limited to, fed batch cell cultures. The methods comprise feeding the cells with D-galactose, preferably with a feeding medium containing D-galactose, preferably daily, to sustain an effective sialylation level of D-galactose in the culture for its duration, thereby increasing sialylation. of the proteins produced. The methods may also comprise at least two temperature changes made during the culture period, in which the temperature is lower at the end of the culture period than at the time of starting cell culture. The cell culture process of the invention involves two or more temperature changes while maintaining high cell viability, and may allow an extended protein production phase.
