abstract An efficient method for the synthesis of taxol, taxol analogs and their intermediates is described. The method involves linking taxol A-ring side chains to baccatin III and synthesizing taxol and taxol analogs with variable A-ring side chain structures. Rapid and efficient esterification of O-protected isoserine and 3-phenylisoserine with N-benzoyloxycarbonyl on 7-O-protected baccatin III, followed by deprotection-acetylation to prepare Taxol, Celphalomannine and various analogs, including photonucleophilic labeled surrogates.
