abstract | An optical system for optical imaging of samples including nanoscale objects is presented. The optical system includes an imaging lens, an illumination source configured to provide excitation light, a detector, and a substrate for supporting the sample. When the sample is applied on the substrate, a sample interface arranged to reflect excitation light is formed between the sample and the first side of the substrate facing the sample. The optical imaging system is arranged such that excitation light is sent into the substrate via the imaging lens and that the detector receives the reference light and scattered light. The reference light includes the portion of the excitation light that is reflected at the sample interface and collected by the imaging lens, while the scattered light includes the portion of the excitation light that is scattered by the nanoscale object and collected by the imaging lens. The optical system is configured to, in response to the excitation light, cause the nanoscale object to be imaged at the detector by optical contrast of the interference pattern between the reference light and the scattered light. The substrate includes an optical coating disposed on the first side of the substrate such that when the sample is applied to the substrate, the sample is in contact with the optical coating. The degree of reflection of the excitation light at the sample interface is such that the optical contrast is greater than that obtained using the sample interface formed without the optical coating. |
