The method includes: clarifying and filtering the cell culture fluid containing recombinant human interleukin-12 protein; performing compound strong anion exchange chromatography on the cell fluid obtained after the pretreatment, and performing compound weak cation exchange chromatography on the harvested protein sample , the further harvested protein samples are subjected to strong cation exchange chromatography, and finally the purified recombinant human interleukin 12 protein is obtained. The present invention only uses three ion exchange chromatography, with few chromatography steps, and the three purification fillers are all pressure-resistant, high flow rate, and high resolution. The purity is as high as more than 99%, the residual amount of host protein is far less than 0.05%, the content of host DNA is less than 0.1ng/15μg, and the content of bacterial endotoxin meets the requirements, which greatly shortens the production time and cost, and is suitable for industrial production.
