abstract | The present application provides methods of purifying nucleic acid-containing samples to obtain isolated nucleic acids in a desired size range, as well as methods of sequencing nucleic acids in a desired size range. The method includes a) mixing the nucleic acid-containing sample with a precipitation buffer in a vessel to provide a precipitation mixture, wherein the precipitation buffer comprises water, buffer, salt, and polyvinylpyrrolidone (PVP) and/or ficoll. The method also includes precipitating the nucleic acid in the above-described precipitation mixture to provide a precipitated nucleic acid fraction and a remaining sample fraction. The precipitated nucleic acid fraction contains predominantly nucleic acid molecules above the selected size cutoff, while the remaining sample fraction contains predominantly nucleic acid molecules below the selected size cutoff. The method also includes separating the precipitated nucleic acid fraction from the remaining sample fraction. The application also provides related compositions and kits. |
