abstract | The invention relates to the technical field of plant RNA extraction, in particular to a method for extracting polysaccharide polyphenols plant RNA by an improved CTAB method. The method includes the following steps: grinding the plant tissue sample to powder, adding the improved extract solution to water bath for lysing and extracting, centrifuging to collect the supernatant, adding an equal volume of chloroform to mix evenly and then standing, centrifuging to collect the supernatant, adding 6M LiCl solution to precipitate RNA , collect the precipitate by centrifugation, add 75% anhydrous ethanol to wash the precipitate, then add DNA digestion solution, dissolve RNA in water bath, and finally add DNase inactivation reagent. The polysaccharide and polyphenolic plant RNA extracted by the improved method of the invention can be used for the construction of a full-length transcriptome library, the extraction success rate can be increased from 64% to 82%, and the library construction success rate can be increased from 92% to 98%. |
