abstract | The invention discloses a system for measuring the content of C 21 steroids in fermentation broth, and a construction method and application thereof. The invention uses pregnenolone as a standard product to prepare a gradient standard solution, and the optimal measurement wavelength is determined through full-band scanning; the orthogonal experimental design is used to determine the optimal measurement conditions for the content determination of C 21 steroids. Under the measurement conditions, a standard curve was established. The optimal measurement wavelength, optimal measurement conditions and standard curve obtained in the construction process constitute the measurement system, which can be applied to the determination of C 21 steroids in the fermentation broth of plant (Bai Shou Wu) endophytes. The endophyte fermentation broth was prepared as a test sample, the absorbance was measured, and the content of C 21 steroids in the sample was obtained by analyzing according to the established standard curve. The method for measuring the content of C 21 steroids not only has simple operation and fast detection speed, but also has good precision, reproducibility, accuracy and stability. |
