abstract |
The present invention provides a sgRNA combination for CRISPANT to specifically knock out irf2bpl gene in zebrafish to construct a zebrafish epilepsy model, the sequences are shown in SEQ ID NO.1, SEQ ID NO.3 and SEQ ID NO.4. In addition to the 7bp fragment on exon 1 of the zebrafish irf2bpl gene, the irf2bpl gene deletion zebrafish was selected to construct the zebrafish epilepsy model. The invention uses three sgRNAs to target the same gene, maximizes the editing efficiency, and performs gene editing more effectively. The present invention can more efficiently and accurately knock out a specific gene in the genome of an organism, and has the advantages of simple manufacture and low cost. The zebrafish epilepsy model provided by the present invention lays a good foundation for further research on the relationship between irf2bpl mutation and epilepsy pathogenesis and screening of antiepileptic drugs. |
