The invention discloses a method for selecting a miRNA molecular marker for sperm motility of Holstein dairy cows, comprising (1) designing specific primers for the miRNA precursor region of Holstein dairy cows, and performing PCR amplification on the genomic DNA of the dairy cow to be tested to obtain an amplification product; ( 2) The PCR product was digested with BspE I restriction endonuclease and electrophoresed, and three genotypes appeared: TT, TC, and CC; (3) The sperm motility of CC was significantly higher than that of TT. The sperm abnormality rate of CC type was significantly lower than that of TT type, so CC type and TC type were selected to save seeds to increase the frequency of C genotype. The method can improve the accuracy of semen quality selection of dairy cows, be used for the selection and breeding of high-quality dairy cows, and provide a breeding tool for strengthening the elite germplasm of Chinese Holstein dairy cows. |
