The invention provides a genetically engineered bacterium producing 3-hydroxypropionic acid, a construction method and application thereof, and belongs to the technical field of bioengineering. In the present invention, genetic engineering and synthetic biology techniques are used to synergistically express a Alcohol dehydrogenase AdhP and succinate semialdehyde dehydrogenase GabD4, and on this basis, through coenzyme engineering to regulate NADH oxidase to improve the coenzyme NAD + circulating supply, a recombinant genetic engineering was obtained; the genetically engineered bacteria can use 1 , 3-propanediol is a substrate, generates 3-hydroxypropionaldehyde under the action of alcohol dehydrogenase, and is then converted into 3-hydroxypropionic acid through aldehyde dehydrogenase; the genetically engineered bacteria are in the aerobic environment without vitamin B 12 . Under these conditions, the self-sufficiency of intracellular coenzymes is completed, and the efficient production of 3-hydroxypropionic acid is realized. |
