abstract The invention discloses a method for detecting melamine in milk based on Ag-COF-COOH substrate surface-enhanced Raman spectroscopy, and belongs to the technical field of Raman spectroscopy. The method for detecting melamine in milk according to the present invention includes the following steps: (1) adding silver nitrate and COF-COOH material into water, heating for incubation, and then adding sodium citrate solution for reduction to obtain Ag-COF-COOH substrate; (2) mix the milk to be tested and acetonitrile evenly, centrifuge to obtain a supernatant; then mix the supernatant with the Ag-COF-COOH substrate of step (1) evenly to obtain a sample solution to be tested; (3) Raman detection is carried out with the sample liquid to be tested in step (2) to obtain a Raman spectrum; the peak intensity at the characteristic peak 702cm -1 ±5cm -1 in the Raman spectrum is brought into the standard curve model, and the melamine in the milk to be tested is obtained concentration. The detection linear range of the method of the present invention is 1-20 μg/L, and the detection limit LOD=0.68 μg/L.
