titleOfInvention A method for establishing mitochondrial unfolded protein reaction system
abstract The invention discloses a method for establishing a mitochondrial unfolded protein reaction system. The method comprises the following steps: 1) according to the molecular formula of G-TPP, synthesizing high-purity TRAP1 inhibitor G-TPP; 2) diluting the G-TPP described in step 1) into a storage solution of 10 mmol/ml in DMSO 3) Cultivate humanized cells with a culture medium containing 10 vol% serum, until the cells are covered more than 90%, add the G-TPP storage solution in step 2) to make the final concentration 10 μmol/ml, and process for 2-6h . This method uses G-TPP, a TRAP1 specific inhibitor, to treat human cell lines, which can interfere with the correct folding of mitochondrial proteins and rapidly induce UPR MT , but does not activate UPR ER , which provides further research on the molecular mechanism of UPR MT and UPR MT . The role in human disease and viral infection provides technical means.
