abstract | The present disclosure relates to methods of enzymatic treatment of double-stranded PCR amplification products for the elimination or minimization of primer dimers in multiplex PCR reactions and for efficient ligation of adaptors. The present disclosure relates to methods and compositions that allow for more efficient amplification of highly multiplexed targets by minimizing laboratory steps, eliminating primer dimers, and increasing the efficiency of adaptor ligation compared to conventional methods, compositions and kits. The disclosed methods use a variety of target-specific primers to specifically and selectively amplify targets in the genome of a subject. The disclosed methods can be used for many downstream procedures and analyses, including DNA sequencing. |
