abstract | The present invention relates to an efficient, sensitive and specific detection method for protein ubiquitination modification in biological samples. The ubiquitination modification detection reagent of the method of the present invention is an artificially synthesized polypeptide (ThUBD) composed of two different ubiquitin-binding domains in series, and the N-terminal contains the tag protein GST, and the GST tag is combined with horseradish peroxide through the GST tag. Enzyme (HRP) or Fluorescein ligation. The detection reagent and method can truly and effectively detect the ubiquitination signal in the formaldehyde-fixed paraffin-embedded tissue sample (FFPE), and realize the fluorescence imaging analysis of the endogenous polyubiquitin chain signal of the biological sample. Compared with the commercial antibody method and the published ubiquitination signal detection method, the inventive method has efficient, concise, sensitive and specific detection performance, and can provide technical means for the detection of ubiquitinated protein regulation in cells and tissues. |
